ABSTRACT

Spodoptera frugiperda (SMITH, 1797) (Lepidoptera: Noctuidae) affects diverse crops of great economic interest, for instance, it can cause severe yield losses in maize, rice and sorghum. In this study, a selection and characterization of Bacillus thuringiensis (BERLINER, 1911) isolates with a high insecticidal activity against S. frugiperda was performed. Fifty-two crystal-forming B. thuringiensis isolates that were identified from 3384 Bacillus-like colonies were examined and screened by PCR for the presence cry genes (cry1, cry1Aa, cry1Ab, cry1Ac, cry1D, cry2 and cry2Ab). Four isolates that showed high toxicity towards S. frugiperda were shown to harbor cry2 genes. The crystals were analyzed by electron microscopy and showed bipyramidal and cuboidal shapes. Furthermore, these four isolates had lethal concentration (LC50) values of 44.5 ng/cm2 (SUFT01), 74.0 ng/cm2 (SUFT02), 89.0 ng/cm2 (SUFT03) and 108 ng/cm2 (SUFT 04) to neonate S. frugiperda larvae. An ultrastructural analysis of midgut cells from S. frugiperda incubated with the SUFT01 spore-crystal complex showed disruptions in cellular integrity and in the microvilli of the midgut columnar cells. The isolates characterized in this work are good candidates for the control of S. frugiperda, and could be used for the formulation of new bioinsecticides.


Spodoptera frugiperda (SMITH, 1797) (Lepidoptera: Noctuidae) afeta diversas culturas de grande interesse econômico, por exemplo, pode causar severas perdas em milho, arroz e sorgo. Neste estudo, foi realizada uma seleção e caracterização de isolados de Bacillus thuringiensis (BERLINER, 1911) com elevada atividade inseticida contra S. frugiperda. Cinquenta e dois isolados formadores de cristal B. thuringiensis que foram identificados a partir de 3384 colônias foram examinados e testados por PCR para a presença dos genes cry (cry1, cry1Aa, cry1Ab, cry1Ac, cry1D, cry2 e cry2Ab). Quatro isolados que apresentaram alta toxicidade contra S. frugiperda foram mostrados para abrigar os genes cry2. Os cristais foram analisados por microscopia eletrônica e mostraram formas bipiramidais e cúbicas. Os valores da concentração letal (CL50) destes quatro isolados foram de 44,5 ng / cm2 (SUFT01), 74,0 ng / cm2 (SUFT02), 89,0 ng / cm2 (SUFT03) e 108 ng / cm2 (suft 04) para larvas recém-eclodidas de S. frugiperda. Uma análise ultra-estrutural das células do intestino médio de S. frugiperda incubadas com complexo esporo-cristal do isolado SUFT01 mostrou rupturas na integridade celular e microvilosidades das células cilíndricas do intestino médio. Neste estudo, o alto nível de atividade inseticida de isolados os torna excelentes candidatos para o controlo de S. frugiperda, e pode proporcionar alternativas no controle destas populações de pragas, bem como a formação de novos bioinsecticidas.

ABSTRACT

Bacillus thuringiensis es un bacilo Gram positivo que durante su fase de esporulación produce unainclusión parasporal, conformada por proteínas Cry con actividad biológica contra insectos-plaga. Gracias a estas proteínas Bacillus thuringiensis presenta toxicidad contra larvas de insectos-plaga de los órdenes Lepidóptera, Coleóptera y Díptera, entre otros. Además es amigable con el medioambiente, razones por la cuales se ha hecho común el uso y desarrollo de productos comerciales y plantas transgénicas a base de toxinas Cry en el sector agrícola. En esta revisión se describirá los aspectos más importantes de Bt y su aplicación como herramienta biotecnológica para el sector agrícola, con una consideración sobre bioensayos usando esta bacteria.


Bacillus thuringiensis (Bt) is a Gram-positive rod-shape bacterium that during its sporulation phase produces a parasporal inclusion formed by Cry proteins having biological activity against pest insects. Because of these proteins, Bt shows toxicity toward pest insect larvae of Lepidoptera, Coleoptera and Diptera orders among others, besides of friendly with the environment, and for this reason Bt has a very common use for commercial products and transgenic plants with Cry protein basis in agriculture. In this review is described the most important Bt factors and their application as biotechnological tool in agrobusiness, considering Bt bioassays.

ABSTRACT

O objetivo deste trabalho foi avaliar a ocorrência do microácaro-da-face-inferior-das-folhas-de-milho Catarhinus tricholaenae Keifer (Acari: Diptilomiopidae) em cultivares transgênicas de milho, contendo as proteínas Cry1F e Cry 1 A(b) e milho não Bt. Durante o período de junho de 2010 a janeiro de 2011, foram coletadas, quinzenalmente, cinco amostras aleatórias de quatro folhas em talhões de milho Bt, contendo a proteína Cry 1F e Cry 1 A(b), e de milho não Bt em áreas experimentais da Embrapa Milho e Sorgo, em Sete Lagoas, MG. As amostras de folhas foram vistoriadas por 15 minutos na região da nervura central, em busca de adultos de C. tricholaenae. Foram registrados 2.930 indivíduos de C. tricholaenae, sendo que 1.114 no milho Bt Cry 1F, 753 em Cry 1 A(b) e 1063 indivíduos em folhas das cultivares não Bt. As maiores abundâncias populacionais médias ocorreram nos meses de novembro e dezembro. Os fatores estágio fenológico das plantas e precipitação afetaram positivamente a abundância de C. tricholaenae. A abundância média do período de coleta de C. tricholaenae foi reduzida pela cultivar de milho contendo a proteína Cry 1 A(b). Esse é o primeiro registro de ácaros sobre cultivares de milho transgênico no Brasil.


The objective of this study was to evaluate the occurrence of "microácaro-da-face-inferior-das-folhas-de-milho" Catarhinus tricholaenae Keifer (Acari: Diptilomiopidae) on transgenic cultivars of corn containing proteins Cry1F and Cry 1 A (b) and non-Bt corn. During the period from June 2010 to January 2011 were collected, every two weeks, five random samples of four leaves in plots of Bt corn containing the protein Cry 1F and Cry 1 A (b) and non-Bt corn in the experimental area of Embrapa Corn and Sorghum, Sete Lagoas, MG. The leaf samples were examined for 15 minutes in the central region of leaf in search of adult C. tricholaenae. We recorded 2930 individuals of C. tricholaenae, 1114 on Bt Cry 1F, 752 on Cry 1 A (b) and 1063 individuals on leaves of non-Bt cultivars. The highest average population abundances occurred in the months of November and December. Factors of plant phenological stage and rainfall positively affected the abundance of C. tricholaenae. The average abundance of the collection period of C. tricholaenae was reduced by cultivar corn containing Cry 1 A (b) protein. This is the first report of mites on the cultivars of transgenic corn in Brazil.

ABSTRACT

Bacillus thuringiensis harbors genes encoding Cry proteins found in chromosomes or plasmids of different sizes (4-150 Mb). Although the smaller plasmids are more abundant in B. thuringiensis, their specific function is unknown. As for the megaplasmids, their main recognized function is to harbor cry genes, although the sequencing of some of these plasmids indicates the occurrence of other important genes. This work used a new protocol for practical and rapid extraction of plasmid DNA in order to characterize the plasmid patterns of Brazilian strains belonging to Embrapa Milho e Sorgo research center B. thuringiensis bank. We tried to further assess the relationship of plasmid patterns with strains belonging to the same serovars and strains causing 100 percent and no mortality to Spodoptera frugiperda (J.E. Smith) larvae. It was possible to characterize 59 strains based on the migration of bands in agarose gel. Strains belonging to the same serovars showed different plasmid sizes (from 1,636 bp to 23,200 bp), with the exception of two strains belonging to serovar galleriae. The strain T09 Bt tolworthi showed a plasmid migration pattern identical to strains belonging to serovar galleriae. Plasmid patterns differed for 46 strains, confirming that this is a useful tool to discriminate specific strains. However, it was not possible to associate the plasmid pattern or the occurrence of particular plasmids with the pathogenicity of a given species towards S. frugiperda larvae.
